The p53 regulatory network
We investigate the cofactors, protein modifications and feedback loops that tune p53 transcriptional output and tumor suppression.
p53 activity is governed by a network of interactions rather than a single on/off switch. The laboratory studies the positive and negative functions of MDM2, the p53 C-terminal regulatory region and the contribution of chromatin context to target-gene selection.
Defining this network reveals points where wild-type p53 may be restored or redirected in cancer cells while also clarifying why seemingly similar mutations produce different outcomes.
